Carrying the Microscope
- ✓ Always use two hands: one firmly holding the arm, and the other supporting the flat base.
- ✓ Keep the cord bundled neatly and sit the microscope away from the desk edge.
- ✓ Clean lenses only with specialized lens paper. Paper towels will scratch the delicate glass!
The Golden Rule
NEVER use the Coarse Adjustment Knob when the microscope is on High Power (40x).
The working distance is extremely small. Using the coarse knob can force the lens directly into the glass slide, shattering both the specimen and damaging the objective lens!
Start on scanning power (4x), center your specimen, focus, and only then rotate to higher power.

How Optics Play Tricks
When light passes through a series of convex lenses in a compound microscope, the image is flipped both horizontally and vertically.
This means:
- An upright letter "e" will appear upside down and backwards: "ë".
- Moving the slide to the left makes the image move to the right.
- Moving the slide up makes the image move down.

Workstation Safety Protocol
Verify safety steps before touching any equipment. Check each item to confirm compliance.
Carry with two hands: support the base and hold the arm. ONLY clean glass surfaces with specialized lens paper. NEVER use the coarse focus knob under medium or high power. Return stage to base position and set scanning lens (4x) first.
Pre-Lab Warm-Up Questions
1. Calculate the total magnification if the ocular is 10x and the objective is 40x.
2. Explain why coarse adjustment knob usage under high power breaks glass slides.
Part 1: The Letter "e" Procedure
- Rotate the objective nosepiece until the scanning power (4x) clicks.
- Mount the letter "e" slide on the stage and secure with stage clips.
- Using coarse focus, raise stage fully; peer through ocular and slowly adjust down to focus.
- Once clear, center the letter, then rotate to 10x power and refocus using ONLY fine adjustment.
- Record and sketch your findings under 40x, 100x, and 400x total magnification on Page 2.
