CDC Scrub Protocol
Stage 1: The Invisible Threat 0:00 – 0:05
Look closely at your hands right now. Are they truly clean?
The average human hand carries over 150 different species of microbes at any given second.
The Detective Challenge:
If dangerous bacteria and viruses are completely invisible to the human eye, how can we prove whether our hands are clean or contaminated?
Microbe Fact
One single gram of soil or dust can contain up to 1 billion bacteria. You touch your face roughly 23 times every hour!
Glow Germ Detectives Investigation Underway
Stage 2: The Chain of Transmission 0:05 – 0:10
How fast does an invisible germ spread?
Our Patient Zero touched a single object coated with invisible glow powder. Watch what happens next:
1
Direct Handshake
Friction transfers thousands of microscopic particles instantly onto a partner's palm.
2
Shared Object
Pencils, door handles, and lab tables become contaminated deposit stations (fomites).
3
Facial Entry Point
Unconscious rubbing of eyes, nose, or mouth transfers pathogens inside the human body.
UV Light Reveal Rule: Glow powder is completely invisible in room lighting, but glows neon under 365–395nm UV radiation!
Stage 3: Simulated Inoculation 0:10 – 0:15
Apply Your Glow Lotion
A
Take exactly one dime-sized drop of glow lotion on your palm.
B
Rub thoroughly: cover palms, backs of hands, finger webs, cuticles, and wrists.
C
Inspect under UV light right now. Notice how entire hands glow brightly!
Lab Safety Check
NEVER look directly into the UV flashlight or point it at someone's eyes. Keep beams pointed down at hands and lab surfaces only.
Hands are now 100% "Infected" Ready for the Washing Experiment
Stage 4: The Controlled Test 0:15 – 0:22
Test Your Handwashing Technique
Each table splits into two investigation squads to test variables:
Squad A The "Quick Splash"
- Rinse under cold water only
- Wash for only 5 seconds
- No soap or light fingertip splash
- Pat dry on paper towel
Squad B The "CDC Protocol"
- Wet with clean running water
- Full soap lather for 20 seconds
- Friction on webs, nails, & thumbs
- Rinse thoroughly & dry cleanly
Prediction: How much glow residue will remain on Squad A vs. Squad B?
Stage 5: Blacklight Forensic Audit 0:22 – 0:26
The UV Reveal: Where Do Germs Hide?
Turn lights off and hold the UV flashlight 6 inches from your hands. Check these four critical hotspots:
Zone 1
Under Nails
Cuticles and finger tips trap 80% of bacteria. Requires deliberate claw scratching against palms!
Zone 2
Between Webs
Quick washing skips the deep skin folds between fingers. Must interlock fingers to scrub!
Zone 3
The Thumbs
Opposable thumbs do the most touching, but palm-to-palm washing completely misses them!
Zone 4
Backs & Wrists
Most quick-washers only rub palm-to-palm, leaving the entire dorsal side untreated.
Record and shade all glowing areas on your Lab Sheet evidence map!
Stage 6: The Master Protocol 0:26 – 0:28
The 20-Second Six-Step Scrub
Happy Birthday song × 2
1
Palms
Palm to palm rubbing
2
Back of Hands
Fingers interlaced over back
3
Interlaced Webs
Palm to palm, fingers woven
4
Backs of Fingers
Knuckles locked in opposite palm
5
Rotational Thumbs
Clasp thumb and twist
6
Fingernails & Wrists
Scrub claw tips circular in palm
Science Fact: Soap molecules have a head that loves water and a tail that latches onto germ oils, pulling them down the drain!
Stage 7: The Debrief & Exit Mission 0:28 – 0:30
Case Closed: Three Forensic Truths
Looks Are Deceiving
Hands that look spotlessly clean under white light can still harbor millions of microbes.
Time + Soap = Friction
A 5-second rinse moves dirt around. A 20-second soap lather chemically destroys germ membranes.
Target The Blindspots
Always scrub thumb wraps, cuticles, and finger webs—the primary zones where pathogens survive.

Blacklight Safety Rule: Never point UV beams into eyes. Inspect hands downward over the lab tray.
PROTOCOL
1 Part 1: The Invisible Transmission Chain (Glow Powder) Contact Spread Test
One "Patient Zero" received a pinch of invisible glow powder on their right palm, then shook hands and passed classroom items across your squad.
Detective Hypothesis:
How many objects or people will show glowing powder under UV light?
Blacklight Evidence Observed:
List 3 specific places glow powder transferred unexpectedly:
A.
B.
C.
2 Part 2: The Wash Variable Experiment (Glow Lotion) Controlled Wash Test
Assigned Wash Squad (Check one):
Squad A: Quick Rinse (5 sec, cold, no soap) Squad B: CDC Protocol (20 sec, soap & scrub)
Pre-Wash Baseline UV Check:
After rubbing the dime-sized glow lotion evenly across both hands, inspect under the UV lamp.
Coverage under visible room light: Appears completely invisible
Coverage under 395nm UV light: 100% Intense Neon Glow
Forensic Prediction: What percentage of glow germ lotion will remain after your wash?
0% (All clean)
1% – 25% (Slight)
26% – 50% (Moderate)
> 50% (Heavy residue)

Strict 30-Minute Minute-by-Minute Pacing Guide
00:00–05:00
Hook & The Invisible Microbe Question
Project Slide 1–2. Ask: "Are your hands clean right now?" Introduce the microscopic nature of viruses and bacteria. Announce that students are now certified forensic detectives.
05:00–10:00
Mission 1: The Glow Powder Transmission Chain
Slide 3. Apply a tiny dab of glow powder to one student's palm secretly or openly. Have them shake hands with one peer, who then touches a pencil and desk. Dim lights and turn on UV light to reveal the rapid spread. Record on Lab Sheet Part 1.
10:00–14:00
Mission 2: Glow Lotion Inoculation & Baseline Check
Slide 4. Distribute 1 dime-sized squirt of glow lotion per student. Students rub it thoroughly all over hands. Use UV lights to verify complete 100% glowing coverage. Assign Squad A vs Squad B.
14:00–21:00
The Controlled Wash Experiment (Sink Rotations)
Slide 5. Squad A does 5s cold water rinse (no soap). Squad B does 20s warm soap scrub. Keep physical line organized. Students dry with paper towels without rubbing excessively.
21:00–26:00
Forensic UV Audit & Evidence Log Table
Slide 6. Dim lights again. Students scan hands with UV light and record residue levels in their Lab Sheet Part 3 table. Complete hotspot checklist.
26:00–30:00
